The number of biologic-based therapeutics is growing rapidly, offering patients improved efficacy but without the ease of oral administration like small molecule therapeutics. Small molecules are typically offered as tablets or capsules that diffuse into the bloodstream through the GI tract. Biologics, however, cannot easily diffuse through the GI tract and may degrade in the stomach or small intestine. As a result, novel drug delivery technologies are needed to enable biologics to be delivered through injection or implant. A common means of delivering drugs is by combining the drug substance with poly(lactic-co-glycolic acid) polymer. PLGA based polymers have been used successfully for the sustained release of biologic drugs but have their limitations. Landec’s drug delivery platform is based on its proprietary Intelimer Polymer® technology and uses PLGAs, Polycarbonates, PEGs, Polycaprolactones as well as Polyacrylates polymerized with fatty groups to yield an optimally bioerodible polymer that can overcome many of these limitations while delivering sustained release of the active ingredient with minimal initial burst.

Injectable and Implantable Drug Delivery for Peptides and Proteins

The flexible processing properties of Landec’s drug delivery platform enable proteins and peptides to be delivered in novel dosage forms that are currently not easily obtained by traditional polymer delivery systems.

Landec’s drug delivery system involves a semi-crystalline polymer at body temperature (the use regime) that exhibits a low melting point and low viscosity properties when heated above the tunable melting point (the processing regime).  The peptide or protein can be incorporated into the polymer matrix a few degrees above the low polymer melting temperature preventing thermal degradation of the biologic active. The biologic active is encapsulated in a semi-crystalline matrix that provides protection for the protein’s secondary and tertiary structures as well as a medium for sustained release.

Oral Drug Delivery

Intelimer Polymer® controlled release systems can be tailored to deliver therapeutics by sustained release or through an external trigger such as pH or temperature. Landec has formulated drug delivery systems incorporating therapeutics with limited aqueous solubility (BCS Class II) by manipulating the hydrophilic/hydrophobic balance of the polymers.
